چکیده

Lipogenesis from propionate-l-Cl4 was first reported by Feller and Feist (2) in slices of liver and adipose tissue. Favarger and Gerlach (3, 4) and Masoro and Porter (5) recently studied the incorporation of propionate-1-V into fatty acids of rat and mouse adipose tissue in viva and in vitro. The incorporation of propionate, compared with that of acetate, was quite low. Favarger and Gerlach (4), on degrading the fatty acids formed from propionate, found labeling to be mainly in pentadecanoic and heptadecanoic acids, with propionate providing the terminal methyl 3-carbon unit. James, Peeters, and Lauryssens (6) found, after perfusion of cow udder with propionate-1-C14, a small amount of labeled oddand even-numbered carbon fatty acids in butterfat. Synthesis of odd-numbered carbon acids from propionate by a cell-free fat-synthesizing enzyme preparation has been reported (7). We have studied propionate metabolism by slices prepared from the mammary glands and livers of lactating and nonlactating rats. In this paper, studies on lipogenesis from, and the oxidation of, V-labeled propionate and succinate by preparations from lactating rats are described.

The Influence of Glucose on Amino Acid Carbon Incorporation into Proteins, Fatty Acids, and Carbon Dioxide by Lactating Rat Mammary Gland Slices.

It has been shown that the capacity of epididymal fat pads (l), diaphragm (2, 3), and liver slices (2) to incorporate amino acid carbon into proteins is lower when those tissues are excised from unfed rather than from fed rats. The addition of glucose to the incubation media containing those tissues from unfed rats increases the incorporation to about the level observed when the same tissues ar...

The relation of glucose oxidation to lipogenesis in mammary tissue.

Earlier studies with rat liver slices indicated that lipogenesis is dependent upon glucose utilization. Thus, liver slices prepared from fasted rats suffer from a lowered capacity to convert added acetate to fatty arids, and this defect in lipogenesis can be repaired by previous administration of glucose (1). hlore recently it was shown that an increase in fatty acid synthesis from acetate can ...

The pentose cycle as a pathway for glucose metabolism in intact lactating dairy cows.

The classical scheme of Embden-Meyerhof has been generally accepted as describing the pathway for glucose metabolism in animals, although recent observations have challenged this tenet. A number of in vitro studies based on the differential conversion of specifically labeled glucose to COZ have indicated the existence of a pathway in addition to the EmbdenMeyerhof pathway for glucose metabolism...

Further observations on the in vitro stimulation by insulin of fat synthesis by lactating mammary gland slices.

The incorporation of [carboxy-14C] acetate into fatty acids by rat-liver slices in vitro was studied by Bloch & Kramer (1948), who showed that the rate of incorporation was increased by pyruvate and still further enhanced if insulin was present as well. The insulin effect was, however, not seen if glucose replaced pyruvate, nor did it occur in acetate alone. Later, Bloch (1948) reported that th...

Alterations in mammary-gland blood flow and glucose metabolism in the lactating rat induced by short-term starvation and refeeding.

Six-hour starvation of lactating rats caused a 75% decrease in mammary-gland lipogenesis. The inhibition of lipogenesis was accompanied by a 45% decrease of blood flow to the mammary gland and a 60% decrease in glucose uptake. Within 2 h of refeeding, lipogenesis and glucose uptake by the gland increased to fed values though blood flow to the gland remained at only 63% of that in the fed state....
